    Investigating plant micro‐remains embedded in dental calculus of the Phoenician inhabitants of Motya (Sicily, Italy)

    Plant records reveal remarkable evidence about past environments and human cultures. Exploiting dental calculus analysis and using a combined approach of microscopy and gas chromatography mass spectrometry, our research outlines dietary ecology and phytomedicinal practices of the ancient community of Motya (Sicily, eight to sixth century BC), one of the most important Phoenician settlements in the Mediterranean basin. Micro‐remains suggest use or consumption of Triticeae cereals, and animal‐derived sources (e.g., milk and aquatic birds). Markers of grape (or wine), herbs, and rhizomes, endemic of Mediterranean latitudes and the East, provide insight into the subsistence of this colony, in terms of foodstuffs and phytotherapeutic products. The application of resins and wood of Gymnosperms for social and cultural purposes is hypothesized through the identification of Pinaceae secondary metabolites and pollen grains. The information hidden in dental calculus discloses the strong human‐plant interaction in Motya’s Phoenician community, in terms of cultural traditions and land use

    Mixture of calcium, magnesium and seaweed affects leaf phytoalexin contents and grape ripening on vines with grapevine leaf stripe disease

    Grapevine leaf stripe disease (GLSD) is a tracheomycosis caused by Phaeomoniella chlamydospora and Phaeoacremonium minimum. Impacts on yields of grapes were correlated with the incidence and severity of GLSD symptoms on vine canopies. In 2012 and 2013, vines in two vineyards were treated with nine applications of a mixture of leaf fertilizers containing calcium, magnesium and seaweed extracts. At different growth stages, leaves were sampled from treated and control plots of healthy, GLSD-asymptomatic, or GLSD-symptomatic vines and contents were measured of the phytoalexins trans-resveratrol, trans-Δ-viniferin, trans-ÎŽ-viniferin and trans-pterostilbene. Grape ripening was also monitored from veraison to harvest during both vintages. The treatments caused significant reductions in canopy symptom expression. Increased phytoalexin contents were measured from ‘fruit set’ to ‘berries developing colour’ stages. Trans-resveratrol peak was recorded in asymptomatic diseased vines at the ‘berries pea-sized’ stage, and trans-Δ-viniferin and trans-ÎŽ-viniferin increased at the ‘berries beginning to touch’ stage, compared to the contents recorded in untreated asymptomatic vines. From ‘berries developing colour’ to harvest, all treated vines had lower amounts of phytoalexins than the control ones. At harvest, treated healthy and symptomatic vines produced berries with similar amounts of total sugars compared to untreated vines. Treated asymptomatic vines produced berries with greater amounts of total sugars compared to the untreated vines. These results indicate that increased phytoalexin content recorded from ‘fruit set’ to ‘berries beginning to touch’ in asymptomatic vines treated with the mineral/seaweed mixture may reduce symptoms of GLSD

    Due to its geographic position and geomorphological configuration, Grotta Romanelli acted as a sediment trap since at least MIS 5. The so-called ‘terre brune’ sequence is a deposit mainly of eolian origin bearing upper Palaeolithic artefacts and fossil remains of vertebrate fauna; it was deposited during the Glacial-Interglacial transition and the Holocene. Sedimentology and mineralogy of this deposit are investigated. The stratigraphic sequence provides a promising archive within which both human and climatic impacts can be studied

    Vineyards containing vines affected with grapevine leaf stripe disease (GLSD), one of the diseases of the esca complex, suffer losses in grape yield and quality every growing season. To examine the relation between GLSD foliar symptoms and levels of phytoalexins in grapevine, phytoalexin levels were monitored in the leaves of symptomatic, asymptomatic/diseased, and healthy grapevine leaves, at various growth stages, in two vineyards in Italy, over four growing seasons. At the same time, the leaf symptoms of the vines at some of those growth stages were recorded in each vineyard and in each growing season. The compounds extracted and identified were: trans-resveratrol, trans-pterostilbene, trans-Δ-viniferin and trans-Ύ-viniferin. The most common phytoalexin found was resveratrol. Amounts of all the phytoalexins were generally greater in symptomatic leaves than in asymptomatic/diseased or healthy leaves. In symptomatic leaves, resveratrol levels were greatest at pre-bunch closure, and peaks in pterostilbene occurred at the same time. Leaves of each category (symptomatic, asymptomatic/diseased, healthy) had lower amounts of these compounds at veraison and generally higher amounts at the stages of harvesting and/or the softening of berries. It seems therefore that the formation and pattern over time of the phytoalexins was linked to the growth stage of the vines. Leaf symptoms never occurred before pre-bunch closure, but became much more common from veraison to harvest. This study provides evidence of a relationship between the levels of phytoalexins, grapevine growth stage, and the seasonal pattern of development of GLSD symptoms

    Who were the miners of Allumiere? A multidisciplinary approach to reconstruct the osteobiography of an Italian worker community

    This research presents an in-depth study of the skeletal remains collected from the archaeological site of Allumiere (15th-16th centuries CE; Rome, Italy). A multidisciplinary approach was used, combining skeletal biology, molecular anthropology and archaeobotany with the aim of reconstructing the osteobiography of the alum miners buried at the site. Since 1460, the area of the Tolfa Mountains was significant for the exploitation of alum which was used for a wide range of purposes in the Middle Ages, ranging from woven production to medical practice. A total of 70 individuals (63 adults and 7 juveniles) were studied. The sex ratio of the community indicated a higher prevalence of males with respect to females. Morphological examination indicated occupational musculoskeletal stress markers, which might reflect the specific phase of alum production that each individual was occupied in. Dietary reconstruction was primarily performed through carbon and nitrogen stable isotope analysis with integration of the results obtained by microscopic, genetic and GC-MS investigations on dental calculus. The diet was omnivorous, indicating a reliance on C3-terrestrial protein and evidence for limited C4 consumption by some individuals. Herbivores, such as sheep and cattle, appear to have contributed to the diet more than pigs and chickens. Consumption of Fagaceae and Poaceae species was predominant; moreover, indicators of Brassicaceae and milk and its derivatives were abundantly recurrent in the population, followed by plant oils and theophylline. Furthermore, the detection of pharmacological alkaloids indicated the knowledge and application of medicinal plants by the community. The novel use of multiple techniques based on cutting-edge technologies has provided a unique window on the lifestyles of individuals from one of the first Italian settlements of alum workers

    Elderly transplant-ineligible (NTE) patients represent the majority of patients affected by multiple myeloma (MM). Elderly patients are a highly heterogeneous population, with large variability in health and functional status. Thus, choosing their optimal treatment is challenging. A wide range of first-line treatments is available, and novel-agent combinations, including monoclonal antibodies (mAbs), have recently entered clinical practice. The combination of the anti-CD38 mAb daratumumab with bortezomib, melphalan and prednisone (Dara-VMP) or lenalidomide and dexamethasone (Dara-Rd) demonstrated impressive advantages in terms of progression-free survival and minimal residual disease negativity, as compared to VMP and Rd, without safety concerns. Another anti-CD38 mAb, isatuximab, is showing encouraging results, and new isatuximab-based combinations might enter clinical practice in the future. Nevertheless, available data come from clinical trials with selected patient populations and, to date, the manageability of these regimens in real-life patients or in frail patients remains unknown. Frailty-tailored treatments, including mAbs, are under evaluation in preliminary studies. In this review, we analyze recently approved mAb-based treatments for NTE newly diagnosed MM patients and new combinations under evaluation, focusing on the efficacy and safety of these regimens and on open issues regarding the choice of therapy for elderly patients

    Negative Modulation of the Angiogenic Cascade Induced by Allosteric Kinesin Eg5 Inhibitors in a Gastric Adenocarcinoma In Vitro Model

    Eg5 is a kinesin essential in bipolar spindle formation, overexpressed in tumours, thus representing a new target in cancer therapy. We aimed at evaluating the anti-cancer activity of Eg5 thiadiazoline inhibitors 2 and 41 on gastric adenocarcinoma cells (AGS), focusing on the modulation of angiogenic signalling. Docking studies confirmed a similar interaction with Eg5 to that of the parent compound K858. Thiadiazolines were also tested in combination with Hesperidin (HSD). Cell cycle analysis reveals a reduction of G1 and S phase percentages when 41 is administered as well as HSD in combination with K858. Western blot reveals Eg5 inhibitors capability to reduce PI3K, p-AKT/Akt and p-Erk/Erk expressions; p-Akt/Akt ratio is even more decreased in HSD+2 sample than the p-Erk/Erk ratio in HSD+41 or K858. VEGF expression is reduced when HSD+2 and HSD+41 are administered with respect to compounds alone, after 72 h. ANGPT2 gene expression increases in cells treated with 41 and HSD+2 compared to K858. The wound-healing assay highlights a reduction in the cut in HSD+2 sample compared to 2 and HSD. Thus, Eg5 inhibitors appear to modulate angiogenic signalling by controlling VEGF activity even better if combined with HSD. Overall, Eg5 inhibitors can represent a promising starting point to develop innovative anti-cancer strategies
